A New Hybrid Genus Amenopsis (Orchidaceae) Derived from the Cross between Amesiella and Phalaenopsis
C.C. Tsai Crops Improvement Division Kaohsiung District Agricultural Research and Extension Station Pingtung County Chinese Taipei
Keywords: intergeneric hybridization, embryo rescue,
Abstract
In order to introduce unique characteristics of the spur structure from the flower of Amesiella into Phalaenopsis germplasm, intergeneric hybridization between Phalaenopsis ‘Sogo Yukidian’ and Amesiella philippinensis were conducted by artificial pollination.
Four months later, the capsules were harvested.
The immature embryos with placenta were manipulated and placed into germination medium.
Approximately 300 intergeneric embryos of each capsule were rescued.
after subculturing for one year in the culture medium and two-week hardiness, intergeneric hybrids were cultivated in greenhouse. After one and half years of cultivation, the first flower of the hybrid bloomed. The new hybrid was named as Amenopsis ‘Kaohsiung Magic’ and registered at the Royal Horticulture Society. Leaf morphology of the hybrid is lanceolate, coriaceus and dark green. The hybrid bears greenish-white flower with black markings on the lip and a short spur at the back of the lip.
The immature embryos were rescued after about 105 days of artificial pollination between Phalaenopsis ‘Sogo Yukidian’ (♀) and Amesiella philippinensis (♂)
The capsule was harvested and immersed in 70% ethanol for 30 s, followed by treatment in 1.25% sodium hypochlorite for 15 min, after that, the capsule was washed with sterilized distilled water three times and dissected under sterile conditions in a laminar flow hood.
Immature embryos with placenta were removed and cultured in medium of 3 g L-1 Hyponex No. 1 fertilizer, supplemented with 2 g L-1 peptone, 7 g L-1 agar, 150 ml L-1 coconut water, 30 g L-1sucrose. The pH of the medium was adjusted to 5.8 with 0.1 N NaOH or HCl before autoclaving.
The immature embryos were incubated at 25±2°C and under a light (an intensity 50 μmol m-2 s -1)/dark cycle of 16/8 hours.
The medium for subculture of the plantlet includes 2.5 g L-1 Hyponex No. 1 fertilizer, supplemented with 2 g L-1 peptone (Difco), 80 g L-1 homogenized banana, 30 g L-1 homogenized potato, 10 g L-1 homogenized onion, 0.1 g L-1 inositol, 20 g L-1 sucrose, 7 g L-1 agar (Merck) and 1 g L-1 of activated charcoal. The pH of the medium was also adjusted to 5.8 with 0.1 N NaOH or HCl before autoclaving.

With embryo rescue technique, various intergeneric plants have been obtained from hybridization of Phalaenopsis and Vanda/Ascocenda, Phalaenopsis and Aerangis/Angraecum, as well as Phalaenopsis and Rhynchostylis. Several novel orchids were obtained
including Asconopsis Kdares Orange Lover, Rhynchonopsis Kdares Perfume Angel, Rhynchonopsis Kdares Perfume Beauty, Rhynchonopsis Kdares Perfume Dancer, Rhynchonopsis Kdares Perfume Pixie, and Rhynchonopsis Kdares Perfume Lemon.
In addition, over sixty novel intergeneric genera were obtained, among which 5 have been raised to flowering and registered as Chouara, Chenara, Amenopsis, Linara and Jungara, respectively .

Development of intergeneric hybrids between wind orchids (Sedirea japonica and Neofinetia falcata) and moth orchids (Phalaenopsis alliances)

Abstract
To develop a new type of orchid with low-temperature tolerance and good fragrance, we generated intergeneric crosses between Korea-native wind orchids and tropical Phalaenopsis. In 2009, 160 cross combinations were made among Sedirea japonica, Phalaenopsis, Doritaenopsis and Neofinetia falcata.
High incompatibility was observed from most of crosses as evidenced by traits such as early dropping of seed pods after pod formation or no seed formation after pod maturation.
A total of 528 hybrid seedlings were obtained from 12 cross combinations. From these hybrid plants, several lines were selected based on flower type, inflorescence, cold tolerance, and fragrance, and finally the three best lines were selected. The selected lines were generally intermediate in both phenotype and genetic characters of the two parents, but tended to be more similar to Phalaenopsis.
